Deutsches Krebsforschungszentrum

Krebsforschung am DKFZ umfasst eine große Anzahl unterschiedlicher Disziplinen beginnend mit der zell- oder molekularbiologischen  Grundlagenforschung bis hin zu den klinischen Studien an Krebspatienten.

Im vergangenen Jahr hat das DKFZ eine neue Einheit zur Wirkstoffentwicklung gegründet, mit der Aufgabe, die Erkenntnisse aus der Grundlagenforschung für die Entwicklung neuer Krebsmedikamente zu nutzen. Dazu wurde ein Portfolio von neuen potentiellen Zielen für Medikamente erstellt, mit dessen Hilfe aussichtsreiche Projekte identifiziert und priorisiert werden für eine Entwicklung in der DKFZ Wirkstoffforschungspipeline. Diese beinhaltet zunächst eine standardisierte Targetvalidierung durch eine eigens dafür gegründete Gruppe (finanziert aus Mitteln des „Deutschen Konsortiums für Translationale Krebsforschung“ (DKTK)) mit anschließender Assayentwicklung und Hochdurchsatzscreen an der DKFZ/EMBL Chemical Biology Core Facility (CBCF). Die CBCF betreibt eine Screening Plattform nach Industriestandard mit einer ausgewählten, chemisch-diversen Bibliothek von 80.000 Substanzen sowie einer Einheit zum molekularen Modellieren für Docking Studien und in silico Screens.

Die identifizierten Hits werden in einer angeschlossenen Chemieeinheit medizinalchemisch zu Leitsubstanzen weiterentwickelt, die in einem großen Panel von Krebszelllinien und normalen Zellen sowie relevanten Tier-Tumormodellen evaluiert werden.

Aktuell entwickeln wir Inhibitoren für eine Phosphatase, eine DNA Methyltransferase, und eine Histon-Deacetylase.

Die weitere Kommerzialisierung von erfolgreichen Projekten wird durch Rahmenverträge mit verschiedenen industriellen Partnern wie Bayer Healthcare erleichtert.
